目的 探讨长链非编码RNA SIX3OS1（LncRNA SIX3OS1）在阿尔茨海默病（AD）患者中的表达水平及其与蛋白生物标志物中的β-淀粉样蛋白1-42（Aβ1-42）、磷酸化tau蛋白217（P-tau 217）、神经丝轻链蛋白（NFL）的关联性。方法 纳入157例AD患者（AD组）和45名体检正常者（对照组）。依据临床痴呆评定量表（CDR）评分将AD患者分为轻度AD组（65例）和中重度AD组（92例）。应用受试者操作特征（ROC）曲线分析LncRNA SIX3OS1联合Aβ1-42、P-tau 217及NFL对AD的预测价值。采用皮尔逊相关系数分析AD患者血清LncRNA SIX3OS1表达水平与Aβ1-42、P-tau 217及NFL的相关性。结果 AD组血清LncRNA SIX3OS1、Aβ1-42、P-tau 217及NFL水平均高于对照组（P<0.001）。中重度AD组血清LncRNA SIX3OS1、Aβ1-42、P-tau 217及NFL水平高于轻度AD组（P<0.001）。ROC曲线分析显示，LncRNA SIX3OS1联合Aβ1-42、P-tau 217及NFL预测AD的曲线下面积为0.977，准确度为95.38%。相关分析显示，AD患者血清LncRNA SIX3OS1表达水平与Aβ1-42、P-tau 217及NFL均呈正相关（P<0.001）。结论 AD患者血清LncRNA SIX3OS1表达水平升高，其高表达与蛋白生物标志物异常相关，LncRNA SIX3OS1联合Aβ1-42、P-tau 217及NFL对AD具有较好的筛查价值。
Objective To investigate the expression level of long non-coding RNA SIX3OS1 （LncRNA SIX3OS1） in patients with Alzheimer's disease （AD） and its correlation with protein biomarkers such as β-amyloid 1-42 （Aβ1-42）， phosphorylated tau at threonine 217 （p-tau217）， and neurofilament light chain （NFL）.Methods A total of 157 patients with AD were enrolled as AD group， and 45 healthy individuals who underwent physical examination were enrolled as control group. According to the Clinical Dementia Rating （CDR） score， the patients with AD were divided into mild AD group （65 patients） and moderate-to-severe AD group （92 patients）. The receiver operating characteristic （ROC） curve was used to assess the value of LncRNA SIX3OS1 combined with Aβ1-42， P-tau 217， and NFL in predicting AD， and a Pearson correlation analysis was used to investigate the correlation of the expression level of serum LncRNA SIX3OS1 with Aβ1-42， P-tau 217， and NFL in AD patients.Results Compared with the control group， the AD group had significantly higher serum levels of LncRNA SIX3OS1， Aβ1-42， P-tau 217， and NFL （P<0.001）. Compared with the mild AD group， the moderate-to-severe AD group had significantly higher serum levels of LncRNA SIX3OS1， Aβ1-42， P-tau 217， and NFL （P<0.001）. The ROC curve analysis showed that LncRNA SIX3OS1 combined with Aβ1-42， P-tau 217， and NFL had an area under the ROC curve of 0.977 in predicting AD， with an accuracy of 95.38%. The correlation analysis showed that the expression level of serum LncRNA SIX3OS1 was positively correlated with Aβ1-42， P-tau 217， and NFL in AD patients （P<0.001）.Conclusions There is an increase in the expression level of serum lncRNA SIX3OS1 in AD patients， which is associated with abnormalities in protein biomarkers， and LncRNA SIX3OS1 combined with Aβ1-42， P-tau 217， and NFL has a good value in AD screening.
